First Report on the Establishment of Callus and Cell Suspension Cultures of Acmella radicans, Total Polyphenol Content and Their Antioxidant Activity
[摘要] ObjectiveTo report for the first time the induction of friable callus from leaf explants and the subsequent establishment of a cell suspension culture (CSC) of Acmella radicans, and to evaluate total phenolic content (TPC), total flavonoid content (TFC), and the antioxidant activity of biomass extracts at different stages of the culture.MethodsCallus induction was evaluated in Murashige and Skoog (MS) solid medium with 2,4-dichlorophenoxyacetic acid (2,4-D) and 6-benzylaminopurine (BAP) at 0.0, 0.5, 1.0, and 2.5 mg/L each. The CSC was established in liquid MS medium with 2.5 mg/L 2,4-D, 0.5 mg/L BAP, polyvinylpyrrolidone, L-glutamine, ascorbic acid, citric acid, and coconut water, with growth kinetics monitored over 18 days. Ethanolic extracts obtained every three days were analyzed for TPC (Folin–Ciocalteu), TFC (AlCl3 colorimetric), and antioxidant activity (ABTS and DPPH).ResultsCallus induction was achieved exclusively with 2,4-D, reaching 100% induction. The highest friability was observed with 2.5 mg/L 2,4-D producing 100% friable callus. Optimized conditions for CSC, biomass followed a sigmoidal growth pattern and peaked at 10.02 ± 0.95 g/L on day 14. TPC and TFC initially decreased, then increased to maximum on day 16 with 18 mg GAE/g DB and 16 mg CATE/g DB, respectively. Antioxidant activity followed the same trend, with ABTS reaching 9.4–9.5 mM TE/g DB and 94.5% inhibition, and DPPH 10.0–10.2 mM TE/g DB with 73.7% inhibition, identifying day 16 as the optimal harvest point in A. radicans cultures.ConclusionA. radicans cell suspension cultures represent a promising in vitro culture system for antioxidant metabolite production and provides a basis for future optimization and scale-up studies.
