ENZYME SURFACE MODIFICATION BY POLYMERS FOR IMPROVED DELIVERY
[摘要] Recent results obtained in the author's laboratory on protein surface modification by polymers, a technique now intensively studied to improve the delivery of protein or drugs, are reviewed with particular regard to: (a) procedures of monomethoxypoly(ethylene glycol), mPEG, activation and their influence on enzyme activity, on the possibility to evaluate the number of mPEG chains bound to the protein, and to follow the biodistribution of the adduct; (b) a method to minimize the loss of enzyme activity towards high molecular weight substrates that usually occurs following polymer binding; (c) pharmacokinetic data of polymer modified superoxide dismutase and its binding to cells; (d) an affinity chromatography procedure to evaluate quantitatively the antibody/mPEG-enzyme recognition; (e)new methods to obtain poly(vinylpyrrolidone) protein adducts.
