BIOCHEMICAL EVIDENCE FOR PRESENCE OF LYSOSOMES IN EPIDERMIS
[摘要] Four biochemical tests have demonstrated the presence of lysosomes in the epidermis of man and of the hairless mouse: presence of acid hydrolases, sedimenta-bility, increased specific activity, structure-linked latency. Epidermal homogenates from man and from the hairless mouse contained 4 lysosomal enzymes: acid phosphatase, cathepsin, [beta]-glucuronidase, and aryl sulfatase. Acid phosphatase activity demonstrated sedimentability of the lysosomal fraction and structure-linked latency. Acid pH, hypo-tonic solution, and freezing and thawing did not solubilize epidermal lysosomes. Triton-X-100 produced time-dependent solubilization (latency) of epidermal lysosomes by 2 methods.
