LIPID METABOLISM IN HUMAN SKIN .I. LIPOGENESIS FROM ACETATE-1-14C
[摘要] A method was developed which demonstrated lipogenesis from acetate-1-14C in skin specimens obtained 5-28 hr. post-mortem as well as in skin specimens stored for several weeks at [long dash]67 degrees C. The method required the incubation of small samples of skin (25-50 mg) with acetate-l-14c in Krebs-Ringer phosphate buffer. After alkaline hydrolysis, the 14c labeled lipids were extracted at pH 1 with dichloromethane, and separated into non-saponi-fiable and acidic fractions by paper chromatography in the system of benzene:methanol:5N NH4OH (2:1:2). The amounts of 14C in the 2 fractions were determined and the amounts incorporated/mg skin were calculated.
