Mesenchymal stem cell pre-treatment with cytokines and hydrogen peroxide modifies their adhesion to cardiac endothelium and murine heart sections AND A preliminary study of the molecular regulation of placental growth factor and vascular endothelial growth factor in murine preadipocytes
[摘要] Project 1. I investigated whether pretreating mesechymal stem cells (MSCs) with inflammatory cytokines or hydrogen peroxide (H\(_2\)O\(_2\)) improves their adhesion to cardiovascular endothelium.Pre-treating mMSCs (passage 7-9) with H\(_2\)O\(_2\) and IFNγ increased their static adhesion to murine cardiac endothelial cells. This effect was lost when passage 10 mMSCs were used. hMSC adhesion to human coronary artery endothelial cells increased with SDF-1α and IL-8 pre-treatment.Adhesion of mMSCs to healthy heart sections was decreased by mMSC pretreatment with H\(_2\)O\(_2\), murine IL-8 and SDF-1α. The species of MSC origin, MSC passage number and the adhesive surface influence the manner in which MSC endothelial adhesion is altered by cytokines. Project 2. Placental growth factor (PlGF) is an attractive anti-obesity target. We aimed to explore the molecular regulation of PlGF and VEGF in murine preadipocytes. Wild-type FoxO1 transfection of murine preadipocytes induced considerable increases in PlGF and VEGF expression. H\(_2\)O\(_2\), VEGF, cAMP and forskolin treatments promoted an increase in preadipocyte FoxO1, VEGF and PlGF expression. Enhanced PlGF expression was not accompanied by an increase PlGF secretion. These preliminary results should be interpreted cautiously and their underlying mechanisms characterised.
