Dysregulation of Protein Phosphorylation/Dephosphorylation in Alzheimer's Disease: A Therapeutic Target
[摘要] Studies during the last two decades have provided new insightsinto the molecular mechanism of Alzheimer's disease (AD). One ofthe milestone findings in AD research was the demonstration thatneurofibrillary degeneration characterized by tau pathology iscentral to the pathogenesis of AD and other tauopathies and thatabnormal hyperphosphorylation of tau is pivotal to neurofibrillarydegeneration. This article reviews the recent research advances intau pathology and the underlying dysregulation of the proteinphosphorylation/dephosphorylation system. An updated model of themechanism of neurofibrillary degeneration is also presented, and apromising therapeutic target to treat AD by correctingdysregulation of protein phosphorylation/dephosphorylation isdiscussed.
