[摘要] Short interfering RNAs (siRNAs) are widely used to bring about RNAinterference (RNAi) in mammalian cells. Numerous siRNAs may bedesigned for any target gene though most of which would beincapable of efficiently inducing mammalian RNAi. Certain highlyfunctional siRNAs designed for knockout of a particular gene mayrender unrelated endogenous genes nonfunctional. These majorbottlenecks should be properly eliminated when RNAi technologiesare employed for any experiment in mammalian functional genomics.This paper thus presents essential notes and findings regardingthe proper choice of siRNA-sequence selection algorithms andweb-based online software systems.
