Study on the Population Genetics of the Great Sturgeon (Huso huso) in the Southern Part of the Caspian Sea Using Microsatellite Loci
[摘要] Fourteensetsofmicrosatellitesprimersweretestedonrandomlyselectedindividualsofgreatsturgeon,H.huso.Fivesetsweredevelopedforlakesturgeon(Afu-19,34,39,54,and68)andsevensetsweredevelopedforAdriaticsturgeon(An-0,1,16,20,40,76,and77)andfnallytwosetsweredevelopedforChinesesturgeon(As-73,and74).ExcepttwosetsofAdriaticsturgeonprimers,An-76,andAn-16,therestofprimersreproduciblyamplifedthebelugasturgeonDNA.Fivepairsofprimersoutof14havebeenselectedandconfrmedtobeusedforseparatingthepopulations.Thoseprimersare;Afu-19,Afu-39,Afu-54fromlakesturgeon;As-73fromChinesesturgeon,andAn-77fromAdriaticsturgeon.ThesamplesfromtheeastregionofthesouthernCaspianSeadidnotamplifyatAfu-19,Afu-39,Afu-54,andAn-77loci.However,samplesfrommiddleandwestregionsoftheCaspianSeadidamplifyatAfu-19,Afu-39,Afu-54,An-77lociindifferentmannersofproducingthebands.SamplesfromallthreeregionsdidamplifyatAs-77locus.However,thebandsatthesamplesfromtheeastregionaredistributeddifferentlyfromthebandsatthesamplesfromthemiddleandwestregions.Statisticalanalysisrevealedthatthenumberofallelesperlocusrangedfrom6to11,andonelocushadatleastsixalleles.Populationdifferentiationcombinationtest(Fisher'smethod)basedonbothgenicdifferentiationandongenotypicdifferentiation(G-based)forthetwosetsofsamplesdemonstratedthattheprobabilitiesarehighlysignifcant.ItmeanspossiblytwogreatsturgeonpopulationsexistinthesouthernpartoftheCaspianSea.
