Growth of isolated palisade cells of Arachis hypogaea in vitro☆
[摘要] Mesophyllwassuccessfullyexcisedundersterileconditionsfromtenkindsofangiospermousleavesandculturedinliquidonashaker.Growtheitheroccurredasspheresofsmall-celledtissueintheliquid,orasmassesofsmallcellsadheringtotheglassabovethemenisciintheflasks.Time-lapseobservationsofasinglepalisadeparenchymacellofArachishypogaeaarepresented;theyshowthat,priortonucleardivision,systrophy(symmetricalaggregationofthechloroplastsaroundthenucleus)occurs.Partitioningoccursinanyplane,andtheresultisasmallcallus,withoutresemblancetoanembryo.ThemostsuitableculturemediumforthesecellsandtissueswasHeller'smacroelementswithspecialmicroelements,sucrose,caseinhydrolyzate,2,4-D,andkinetin.Greatlystimulatedgrowthwasachievedbyadditionof0.1mmoleofNH4Clperliter.Continuousculturescouldbeattainedbytransfersatappropriateintervalsofundividedordividedspheresoftissuesderivedfromthemesophyllinocula.Ontransferofthespheresoftissuetoagarmedium,themodifiedHeller'smediumdidnotpermitsurvival.Linsmaier-Skoog'smediumprovedadequatehere,andalarge-celledcalluswasproduced,andcouldbeindefinitelytransferred.Theexcision,growth,anddevelopmentincultureofthehighlydifferentiatedpalisadeparenchymacell,asaprimeexampleofamaturelivingcell,arediscussedinrelationtopreviousresults.
