Permanent effects of estradiol on cellular metabolism of the developing mouse vagina☆
[摘要] Persistent,hormone-independentvaginalproliferationisproducedwhenestradiolisgiventomiceduringadiscreteperiodfollowingbirth.SynthesisofDNA,RNA,andproteininthealteredvaginaehasbeeninvestigated.Anincreasedrateofcellmultiplicationinvaginaefromadultanimals,givenestradiolinthenewbornperiod,isindicatedbyincreasedincorporationofthymidine-3HintoDNA.Incorporationoflabeledaminoacidsintovaginalproteinisaccelerated,asisincorporationofuridine-3HintototalorganRNA.VaginalRNA,labeledinvivowith32P,wasfractionatedbyextractionatthreetemperatures.Theresultsofthermalfractionationsuggestthatthepermanentestradiol-inducedchangesinvaginaeareaccompaniedbyselectiveeffectsonsomeRNAspecies,eitherintermsofrelativeratesofsynthesisorinqualitativedifferencesofRNAmoieties.Vaginaefrommice,givenestradiolinthenewbornperiod,retainselectiveuptakeandprolongedretentionof3H-labeledestradiolandthecapacitytorespondtoexogenousestradiolwithincreasedincorporationoflabeledaminoacidsintoprotein.Thevaginalepitheliumremainscapableofbothmucinandkeratinproduction.Thesestudiesdemonstratethatpersistentalterationsofcellularmetabolismaccompanythemorphologicalsequelaeseeninvaginaefrommicegivenestradiolduringadefinedneonatalperiod.RatesofsynthesisofDNA,RNAandproteinarepermanentlyaltered,whilesomenormalcharacteristicsandhormone-specificresponsesofvaginaeareunaffected.
