Effects of collagenase on developing epithelia in vitro: Lung, ureteric bud, and pancreas☆
[摘要] Culturedepitheliafromembryoniclungs,uretericbuds,andpancreasweretreatedwithcollagenase.Lungmorphogenesisstoppedanduretericbudslosttheircharacteristicshapeaftersuchenzymetreatment.Ifmesodermwassuppliedtotheculturesaftertreatment,lungdevelopmentcontinuedandsomeoftheuretericbudsmaintainedtheirshape.Pancreaticepitheliawereunaffectedbyenzymetreatmentandformedexocrineacinianddifferentiatedcellseveninthepresenceofcollagenase.Electronmicrographsoftreatedlungtissuesshowthatthebasementlaminaandothernearbyextracellularmaterialsareremovedbycollagenase.Theresultsimplythatsomeformofcollagenisanessentialcontributionofmesodermtoepitheliumincaseswhereepithelialbranchingisoccurring,butnotwhereaciniareforming.Theformerprocessiscorrelatedwithhighmesodermspecificityrequirements.Thedatasuggestthatcollagenisessentialforstabilizationofepithelialmorphology;noevidencedemonstratesanyinductivepropertiesforcollageninthesesystems.
