Studies on intercellular adhesive selectivity☆☆☆
[摘要] Theprobabilitythattwocellswillformandmaintainanadhesionaftercollisionoccursinsuspensionisassumedtoindicatetheadhesivestabilitybetweenthecellsinquestion.Unlabeledcellaggregatesarecirculatedintritiatedthymidine-labeledcellsuspensionswhereallextraneousfactorswhichmighthaveaffectedcollisionareheldconstant.Aftercirculating,theaggregatesarecollected,sectionedandexaminedradioautographicallyforlabeledcells.Experimentsarecarriedoutwithcellsandaggregatesfromchickenembryoneuralretinae,livers,sterna,heartventriclesandpectoralmusclesandwithmouseembryoneuralretinaeandheartventricles.Isotypicaggregates(sametissuetypeasthecellsinthesuspension)alwayscollectmorelabeledcellsthansimilar-sizedheterotypicaggregates(differenttissuetype).Collectionsoflabeledchickenembryolivercellsbyunlabeledliveraggregatesisdecreasedbylowtemperatures(0°and22°C)andbypuromycinbutnotbyactinomycinD.“Conditionedâ€mediumincreasesthecollectionandcounteractstheinhibitoryeffectsoflowtemperaturesandpuromycin.Somedegreeofadhesiveselectivityisshownbyneuralretinaandlivercellsandaggregateswhichwerekilledwithglutaraldehyde.Becausethepresenceofunlabeledcellsaltersthecollectionoflabeledcellsofanothertypebyiso-andheterotypicaggregates,itisconcludedthatnonselectiveadhesionsoccurredbetweenfreshlydissociatedcellsofdifferenttypesmorereadilythanbetweencellsandheterotypicaggregates.Ahypothesisisdescribedwhichassumesthatthereareatleasttwodifferentmolecularmoietiesonthecellsurfacewhichareresponsibleforintercellularadhesion.Oneofthesecausesselectiveandtheothernonselectiveadhesions.Experimentstotestpredictionsofthehypothesisarediscussed.
