Tcp-1 gene is not responsible for the maternal lethality effect of Thp mutation in mice
[摘要] EmbryosreceivingeithertheThporthetwLub2mutationfromtheirmotherdieduringgestation.Incontrast,heterozygousembryosreceivingthemutantchromosomefromthefatherarecompletelyviable.InbothThpandtwLub2mutants,onechromosome17carriesadeletion,whichsuggeststheexistenceofadiscretematernallethalityeffectlocus(Tme).IntheregionwhosedeletionisresponsiblefortheThportwLub2mutation,asinglegenehasbeenclonedsofar,theTcp-1gene.Inthepresentstudy,weexaminedtheexpressionofTcp-1geneinmutantembryoscarryingeitheramaternalorapaternalThpchromosometotestwhetherthisgenecouldbeinvolvedintheTmeeffect.WefoundthatthelevelsofTcp-1-specifictranscriptsweresimilarinbothmutantembryosfromreciprocalcrossesandcorrespondedtohalfthelevelsfoundinthenormallittermates.Inaddition,thepaternalandmaternalTcp-1alleleshadidenticalmethylationpatterns.TheseresultsindicatethatTcp-1isnotresponsibleforthematernallethalityeffect,andthereforeisnotlocatedattheTmelocus.
