Effects of thyroxine and low oxygen tension on chondrogenic expression in cell culture☆
[摘要] Theeffectsofthyroxineandlowoxygentensiononembryonicchickchondrocytesincellculturewereinvestigated.Theassaysforchondrogenicexpressioninvolved(1)morphologicalandhistologicalobservationsforaresemblancebetweentheculturedcellsandhyalinecartilageinvivo,(2)measurementsoftheratesofpolysaccharidesynthesisbymonolayers,and(3)analysisofthepolysaccharidesforthepresenceofchondroitinsulfate.Thyroxine(10−6M)wasmarkedlystimulatorytobothmonolayersandclones;adelayedadditionofthehormonetoclonescausedtheconversionsofsomenon-CMC'sintoCMC's.Lowoxygentensionstimulatedtherateofchondroitinsulfatesynthesisbymonolayersbuthadnoapparenteffectonclones.Theeffectsoflowoxygentension,whichwerereversible,wereapparentonlyinthepresenceofaddedthyroxine.Relativelyshort-termexposurestoBUdR(10μg/ml)stronglyinhibitedchondrogenicexpression,eveninthepresenceofthyroxineandlowoxygentension.Theresultssupportpreviousreportsthatthyroxineandlowoxygentensionmayplayaphysiologicalroleinchondrogenesisinvivo.
