Disturbances in developmental pathways leading to a neurological disorder of genetic origin, “leaner,” in mice☆
[摘要] Alinkagerelationshipbetweentheleanergene,la,andesterasegene,El,wasdescribed.Therecombinationfrequencybetweenthesegeneswasfoundtobe3.84%.Developmentalpathwaysleadingtoneurologicaldisordersintheleanermutantwereinvestigated.Itwasfoundthatthethymidineuptakebythebrain,liverandspleenofthemutantmicewassignificantlylowerthanthatoftheirnormalsibs.Therewasagoodcorrelationbetweenthelowerthymidineuptakeandthereducednumberofmitoticfiguresintheliverandcerebellum.Reducedthymidineuptakewasfoundtooccurbetweenday5andday6ofpostnatalage;itpersistedaslongastherewasasufficientamountofDNAsynthesis(orcelldivision).ItwasshownthattherewasafasterbreakdownofDNAsynthesizedinthemutants,butthatthisfasterbreakdownwasanirregularevent,occurringinassociationwiththelatestageoftheaffliction.ItwaspointedoutthatthefasterbreakdownofDNAwasnotthebasisfortheneurologicaldisorders.Aloweruridineandleucineuptakewasoccasionallyobservedinthemutantorgansatthelatestageofaffliction.Thiswaspointedoutasbeingassociatedwiththeatrophyoftheorgans.ThelevelofRNAandproteinshowedasimilartendency.Itwasshownthatadevelopmentallaginthetotalnitrogenlevelofthemutantswasnotaconsistentphenomenon,andthatthislagwasindependentfromthebehavioralabnormalities.ItwasshownthatthemetabolismofRNA,protein,andesteraseinthemutantorganswasrelativelynormalduringtheearlystageofafflictioninspiteofthedrasticreductionintherateofDNAsynthesisandcelldivision.
