In vitro development of the regenerating lens☆
[摘要] ThedevelopmentinvitrooftheregeneratinglensatsuccessivetimeintervalsafterlensremovalwasstudiedintheadultnewtTriturusviridescens.Regenerateswereculturedusinganagarsubstrateinwhichasyntheticmediumwithserumsupplementwasincorporated.Explantswereincubatedat25°C,5%CO2,and85%relativelyhumidity,andculturedfor5,10,and15days.RegenerationstagesItoII,inwhichnuclearactivationanddepigmentationarestartingtooccur,continuetoextrudetheirpigmentgranulesandgiverisetoapopulationofcompletelydepigmentedcells.Afterdepigmentationhasoccurredinvivoinsomecellsofthedorsaliris(stagesIIItoIV),adepigmentedcellpopulationisproducedcapableoflimitedproliferationbutincapableoflensfiberdifferentiation.Whencellelongationhasoccurredinthedepigmentedcellsoftheinnerwalloftheregeneratinglens(stagesVandonward),explantsarecapableofdevelopinglensfibersinvitro.TheseresultsarediscussedandrelatedtotheavailableinformationonRNAandproteinsynthesisinthelensregeneratingsystem.
