Cell surface and cytoskeletal changes associated with epidermal stratification and differentiation in organ cultures of embryonic human skin☆
[摘要] Embryonicandfetalhumanepidermisdifferentiatesinorgancultureinanagedependent,thoughaccelerated,manner.Theolderthespecimenthelesstimeisrequiredforepidermaldifferentiation.Morphologicalmarkersofepidermaldifferentiation,includingthedifferentepidermalstrata,keratohyalingranules,lamellargranules,andcornifiedcellenvelopes,areformedinamannerthatisfaithfultodevelopmentinvivo.Thehighmolecularweight,“differentiationspecific”(67and56.5kDa)keratinsarealsoexpressedinthesecultures,evenintheabsenceofmorphologicalevidenceforkeratinization.Unstratified,embryonicepidermiswasfoundtostratifyovernightinculture.Thetimecourseofcellsurfacechanges,detectedbylectinbinding,andcytoskeletalchanges,detectedbyexpressionofthehighmolecularweightkeratins,wasfollowedinthesecultures.Peanutagglutinin(PNA)bindingsitesappearedovernightinculturecoincidentlywithepidermalstratificationwhileexpressionofthe67and56.5kDakeratinswasnotdetecteduntilthethirddayofculture.Thepossiblesignificanceoftheseresultsisdiscussed.
