[3H]acetylcholine synthesis in cultured ciliary ganglion neurons: Effects of myotube membranes☆
[摘要] Avianciliaryganglionneuronsincellculturewereexaminedforthecapacitytosynthesizeacetylcholine(ACh)fromtheexogenouslysuppliedprecursor,choline.RelevantkineticparametersoftheAChsyntheticsysteminculturedneuronswerefoundtobevirtuallythesameasthoseoftheganglionicterminalsintheintactiris.Neuronswereculturedinthepresenceofandallowedtoinnervatepectoralmuscle;thisresultsinanenhancedcapacityforAChsynthesis.Inparticular,theabilitytoincreaseAChsynthesisupondemandafterstimulationisaffectedbyinteractionwiththetarget.Thiseffectisshowntobeanaccelerationofthematurationoftheculturedneurons.Lysedandwashedmembraneremnantsofthemuscletargetwereabletoduplicate,inpart,thiseffectoflivetargettissueonneuronaltransmittermetabolism.Culturemediumconditionedbymuscle,andbythemembraneremnantsofmuscle,waswithoutsignificanteffect.Thus,substancessecretedintothemediumdonotplayamajorroleinthisinteraction.Neuronsculturedwitheithermuscleormusclemembraneremnantsformedlarge,elongatestructuresonthetargetmembranesurface.Thesewerenotseenintheabsenceofthetargetatthetimesexamined.Thismorphologicaldifferenceinterminal-likestructuresmayparallelthedevelopmentalincreasesinsizeandvesicularcontentofciliaryganglionnerveterminalsinthechickiris,andmayrelatetotheincreasedAChsyntheticactivity.Theresultssuggestthatdirectcontactwithanappropriatetargetmembranehasaprofound,retrogradeinfluenceuponneuronalmetabolicandmorphologicalmaturation.
