Experimentally induced changes in fructose-1,6-diphosphatase activity in embryonic chick liver and heart cells in vitro☆
[摘要] Fructose-1,6-diphosphataseactivityinliverandheartofchickembryoswasassayedinfreshlyisolatedtissue,aftercultivationofthecellsinmonolayers,incellaggregates,intissuefragmentcultures,andingraftsonthechorioallantoicmembrane.Intheliver,specificactivityofthisenzymewashighin6-dayembryos,decreasedrapidlyuntilthefourteenthday,thendecreasedslightlyuntilafterhatching.Intheheart,specificactivitywaslowthroughoutembryonicdevelopmentandafterhatching.Inmonolayerculturesofdissociatedlivercellsfrom8-dayembryosspecificactivitydecreasedrapidlyafter2days,andgraduallyduringfurthercultivation.Inmonolayer-culturedheartcellsspecificactivitywasmaintainedataninitiallowerlevel.AfterIdayofcultivationfibroblast-likecellsandroundcellswereobservedinmonolayerculturesofliverandheartcells.Afterseveraldaysinmonolayercultures,fibroblast-likecellspredominatedoverroundcells.Inaggregatesofdissociatedlivercellsspecificactivitydeclinedrapidlyafter2days,thenincreasedgraduallytoneartheinitiallevels.Inaggregatesofheartcellsspecificactivitydeclinedinthefirst2or3days,thenrosegraduallytoalevelabovethatfoundintheintacthearttissueinvivo.Inaggregatesofliverandofheartcells,thecharacteristichistologicalpatternsofliverandhearttissueswerereconstructed.Infragmentculturesoflivertissue,specificactivityoftheenzymedecreasedrapidlyafter2days,andgraduallyduringfurthercultivation.Infragmentculturesofheart,specificactivitydecreasedafter2days,thenrosetoahighlevel.Ingraftsoflivertissuetothechorioallantoicmembranetherewasagradualdeclineinthespecificactivityoftheenzyme;ingraftsofhearttissuespecificactivityroseduring6daysofincubation.Histologicalobservationsonliverfragmentsincultureandinchorioallantoicgraftsshowedthattheirparenchymalstructurewasretainedfor6days.Inheartfragmentsincultureandinchorioallantoicgraftsthemyocardialstructurewasretainedandabundantmyofibrilswerepresent.
