A reexamination of heart muscle differentiation in the postnodal piece of chick blastoderm mediated by exogenous RNA

[摘要] PreviousfindingsonexogenousRNA-inducedheartmuscledifferentiationinthenon-heartformingculturedexplantsofthechickblastoderm,thepostnodalpieces,werereexamined.Someofthechangesthatcharacterizedthetransitioninthehosttissueswere:(i)theformationofhighlyorderedmyofibrils;(ii)theappearanceofcharacteristiccytoplasmicglycogenparticles;(iii)a2.5-and3.5-foldincreaseinactinandmyosin-likepolypetidesrespectively;(iv)anincreaseinacetylcholinesteraseactivity;and(v)theacquisitionofspontaneousandrhythmicpulsations.Thesespecificchangesappearedonlyinthoseexplantsthatreceivedapoly(A)-containingRNAfractionobtainedfromthe16-day-oldchickembryonicheart.NeithersyntheticpolynucleotidesnoravarietyofRNAfromseveralsourcescouldreplacetheRNAfromchickembryonicheartasaninducerofheartmuscledifferentiation.
