Clusters of acetylcholine receptors located at identified nerve-muscle synapses in vitro☆
[摘要] Nerve-musclesynapsesformwithin24hrafterfragmentsofembryonicchickspinalcordareplacedinestablishedmusclecultures.Nerveprocess-musclefibercontactswerevisualizedwithinterferencecontrastoptics,andsitesoftransmitterreleasewereidentifiedbyextracellularstimulationofoverlyingneuritesinthepresenceoftetrodotoxin.Theresolutionofthisfocaldepolarizationtechniqueislessthan5μm,andresultsindicatethatsynapsesarerestrictedtoshortsegmentsalongunmyelinatedmotoraxons.Thedistributionofacetylcholine(ACh)receptorsoninnervatedmyotubeswasdeterminedbymicrointophoresisofACh.Clustersofreceptors,signaledbysharprelativepeaksofAChsensitivity,werelocatedexactlyatsitesoftransmitterrelease.Thedensityofreceptorswasnotincreasedallalongthelengthofmotoraxon-musclecontacts.
