Interference of tooth differentiation with interposed filters☆
[摘要] TheeffectofinterposedNucleporefiltersontheepithelio-mesenchymalinteractioninembryonicmousetoothwasstudied.Filterswithporesizesof0.6and0.2μmalloweddifferentiationofodontoblastsandameloblastsinthebell-stagetoothgerm.Thisdifferentiationprogressedmorerapidlywhenthe0.6-μmporesizefilterwasused.Nucleporefilterswith0.1-μmporespreventeddifferentiation.Electronmicroscopicexaminationrevealedpenetrationofcellprocessesintothefilterpores.Cytoplasmicmaterialcouldbeseeninthe0.6-μmpore-sizefilterwithin3daysofcultivation,whereas,inthe0.2-μmfilterpores,penetrationwasslight.After6daysofcultivation,cytoplasmicmaterialwasfoundatalllevelsofthe0.2-μmpore-sizefilter,butnotinthechannelsofthe0.1-μmpore-sizefilters,preventingdifferentiation.Itisconcludedthatthe0.1-μmpore-sizefilterblockstoothdevelopmentatthelevelofmesenchymalcelldifferentiationintoodontoblasts.Itissuggestedthatthisdifferentiationrequiresacloseassociationbetweentheinteractingmesenchymalandepithelialcells.
