Mitochondrial DNA synthesis in large and mature oocytes of Xenopus laevis
[摘要] DeoxynucleosidesareincorporatedintomitochondrialDNA(mtDNA)oflargeoocytes;therateofincorporationisabout2%ofthemtDNAamountper24hr.Whenoocyteshavebeeninducedtomatureinvitrowithhumanchorionicgonadotropin(HCG),uptakeandactualincorporationofthymidinedecrease,althoughphosphorylationisenhanced.AnexaminationofmtDNAreplicationshowsthatHCGtreatmentinducesanincreaseintherelativesynthesisofE-strandsandanaccumulationofD-loops.Asimilareffectisobtainedbyethidiumbromidetreatment.Thus,gonadotropinappearstodelayE-strandelongationandtosynchronizemtDNAmoleculesatthebeginingoftheirreplicationcycle.
