The cytoplasmic distribution and characterization of poly(A)+ RNA in oocytes and embryos of Drosophila☆
[摘要] Usingthepresenceofpoly(A)tractsasamarkerformRNA,wehaveexaminedthedistributionofthisclassofRNAbetweenpolysomesandfreeRNPparticles.Thishasbeendoneinmatureoocytesandinembryosagedforvarioustimesfromfertilizationthroughtohatchingofalarva.Theproportionofribosomesthatareinpolysomestothosethatarenothasbeencalculated.Inmatureoocytes,58%ofthepoly(A)+RNAand72%oftheribosomesarenotinpolysomes.By1hr,thisdropsto51%ofthepoly(A)+RNAand48%oftheribosomes.By7hr,aplateauisreached:30%ofeacharenotinpolysomes.Thepoly(A)+RNAinthecytoplasmofoocytesand1-hrembryosisfoundinparticleswithanaveragesizeof50Sandarangeof30–70S.Thepoly(A)+RNArangesinsizefrom7to40S,withanaveragesizeof22S.ThepolyAfromthisRNAis50–200nucleotideslongwithanaverageof115nucleotides.Thesedatahaveallowedustocalculatethat1–2%ofthetotalRNAispoly(A)+RNA.
