In vitro determination and differentiation of mouse teratocarcinoma toward myocardiogenesis
[摘要] Pluripotentialmouseteratocarcinomacellsinembryoidbodieswereexplantedinvitroafterwhichvariousmodesofdifferentiationoccurred.Afterabout5daysinculture,1–4%ofthepiled-upcoloniesexhibitedsynchronizedrhythmicalcontractionwithaconstantfrequencyrangingfrom50to70contractions/min,irrespectiveofthecolonysize.Thisphenomenoncouldbemaintainedfor25daysunderourcultureconditions.Ultrastructuralstudiesoftherhythmicallycontractingcoloniesrevealeddeterminationanddifferentiationofembryonalcarcinomacellsexclusivelytowardmyocardiogenesis,andthedevelopmentalcellularstageswereobserved.Myocardialdifferentiationwascharacterizedbythedevelopmentalstagesoffibrillogenesis,accompaniedbytypicalspecializationofthemyocardial-membranecomplex.Thismyocardialdifferentiationbearsaclosesimilaritytothenormalembryonicmyocardiogenesisof8-to18-daymouseembryoheart.
