Collagen synthesis by normal and bromodeoxyuridine-modulated cells in myogenic culture☆
[摘要] Connectivetissueappearstoplayanimportantroleinthedevelopment,function,andpathologyofvertebrateskeletalmuscle.Inordertoanalyzethesourceoftheextracellularmatrixindevelopingmuscle,hydroxyprolineassaysandfine-structureanalysesweremadeonclonalculturesderivedfromembryonicmusculature.Thecellsinfibroblastculturessynthesizedandaccumulatedanextensiveextracellularmatrixofnonstriatedreticular-likemicrofibrils.Inmyoblastcultures,noextracellularmatrixwasevidentbyelectronmicroscopy,althoughhydroxyprolineassaysdemonstratedthatthecellssynthesizedandreleasedasolublecollagenousproteinintothemedium.Whengrownwithfibroblasts,multinucleatedmyofibersexhibitedacompletebasementmembraneconsistingofbothbasalandreticularlaminae.Bromodeoxyuridine-treatedmyoblasts,inadditiontoexhibitingmorphologicalandcytologicalalterations,synthesizedandelaboratedanextracellularmatrixidenticaltothatinfibroblastcultures.Thepotentialforasimilaralterationor“modulation”ofmyoblastexpressioninsituisdiscussedinregardtoitspossiblerelationtofibrosisinmusculardystrophy.
