Changes in protein phosphorylation accompanying maturation of Xenopus laevis oocytes☆☆☆
[摘要] Proteinphosphorylationhasbeenmeasuredafterinjectionof[32P]phosphateintooocytesofXenopuslaevisundergoingprogesterone-inducedmeioticmaturation.Asoocytesmature,thereisaburstofnonyolkproteinphosphorylationseveralhoursafterprogesteroneexposureandshortlybeforegerminalvesiclebreakdown(GVBD).ThisburstisnotduetochangesinthespecificactivityofthephosphateorATPpool.Enucleatedoocytesexposedtoprogesteronealsoexperiencetheburst,indicatingthecytoplasmiclocationofphosphoproteinformation.Whenanoocytereceivesaninjectionofcytoplasmcontainingthematuration-promotingfactor(MPF),aburstofproteinphosphorylationoccursimmediately,andGVBDoccursshortlythereafter,eveninthepresenceofcycloheximide.Underavarietyofconditionspromotingorblockingmaturation,oocyteswhichundergoGVBDaretheonlyonestohaveexperiencedthephosphorylationburst.TheresultssuggestthattheproteinphosphorylationburstisanecessarystepinthemechanismbywhichMPFpromotesGVBD.
