Factors involved in the stimulation of parasympathetic nerve outgrowth☆
[摘要] Nervegrowthfromthemouseparasympatheticsubmandibularganglionisstimulatedbythedevelopingtargetepithelium.Toinvestigatethenatureofthistrophicinfluence,homogenatesofsalivaryglands,gland-conditionedmedium,andformalin-fixedglandswereassayedforabilitytoelicitparasympatheticaxonextensionintissueculture.Neitherhomogenatesnorconditionedmediumstimulatedaxonoutgrowthfromsubmandibularganglia.However,whengangliawereaddedtoglandsinwhichproteinsynthesisandcellmovementhadbeenhaltedbyformalinfixation,stimulationofoutgrowthintothetissuewasobserved.Stimulationofaxonalgrowthoccurredafterhyaluronidaseandcollagenasetreatmentoftheglands,butnotaftertreatmentwithproteasesorwithheat.Moreover,prolongedformalinfixationdestroyedtheglandularabilitytoelicitaxonelongation.Intactgangliaculturedwithwholelivesubmandibularglandsinthepresenceoflowlevelsofhyaluronidaseorcollagenaseshowedextensiveaxonoutgrowthdespitedisruptionofthenormalmorphogeneticpatternofbothepitheliumandaxons.Ourresultssuggestthatstimulationofaxonoutgrowthdoesnotdirectlydependontheconcomitantmetabolicormorphogeneticactivityoftheepithelium,butiscausedbysomeepithelialproduct,probablyaprotein.
