Appearance and persistence of maternal RNA sequences in sea urchin development☆
[摘要] ThispaperdealswiththerelationshipbetweenthesinglecopytranscriptsrepresentedinmatureoocytesoftheseaurchinandtheRNAsequencespresentinimmatureoocytesandembryos.WetermtheoocytetranscriptsfromsinglecopyDNAthematernalsinglecopysequenceset.Asinglecopy[3H]DNAfraction([3H]oDNA)enrichedforsequencescomplementarytothematernalsinglecopysequencesetwaspreparedandreactedwiththedifferentRNApreparations.ThecomplexityofthematureoocyteRNAisestimatedtobe37×106nucleotides.Atkinetictermination,[3H]oDNAreactedwiththepolysomalmRNAof16-cellembryosto73%ofthereactionwithmatureoocyteRNA,indicatingthat27×106nucleotidesofthematernalsequencesetarepresent.WithblastulamRNAthereactionequalsabout56%,acomplexityof21×106nucleotides;withgastrulamRNA,53%,acomplexityof19×106nucleotides.Therelativeamountofhybridizationof[3H]oDNAwas100%withcytoplasmicRNAofthe16-cellstageandbecameprogressivelylesswiththecytoplasmicRNAsoflaterstages.ThetotalRNAofimmatureoocyteswasfoundtoincludeabout26×106nucleotidesofthematernalsequenceset.Resultsoftheseexperimentsarediscussed,andaninterpretationofthepatternofutilizationofstructuralgenesduringoocyteandembryodevelopmentissuggested.
