Development of the embryonic mammalian pancreas: The relationship between morphogenesis and cytodifferentiation☆☆☆
[摘要] Thedegreetowhichcytodifferentiationisobligatorilycoupledtoongoingmorphogenesishasbeenanalyzedinthedevelopingmousepancreassystem.Protodifferentiatedpancreaticepitheliaareisolatedfrominvestingmesodermandareculturedonplasticsubstrata.Within24hr,theepitheliaspreadintotwo-dimensionalmonolayers.Undertheseconditions,acinarformationfailstotakeplace.Furthermore,thecellsdonotundergomitosis,anobservationconsistentwiththeabsenceofmesenchymalcellsormesenchymalfactorfromthesecultures.Inspiteoftheabsenceofmorphogenesis,cytodifferentiationtakesplaceonschedule.Thus,over5–7daysofculture,amylasespecificactivityrises100-fold,andzymogengranulesappearinthecells.Theresultsarguethattheregulationofexocrinecytodifferentiationisindependentofongoingmorphogenesis.
