Embryonic tissues as elasticoviscous liquids: II. Direct evidence for cell slippage in centrifuged aggregates
[摘要] Certainembryoniccellaggregatesdisplaybothsolid-likeandliquid-likepropertiesinorganculture.Whencentrifugedagainstsolidsubstrata,theseaggregatesundergosuddeninitialdeformationsfollowedbymoregradualshapechanges.Thus,theyareeithercompoundviscoelasticsolids(inwhichcellsfirststretchrapidly,thenslowly)orelasticoviscousliquids(inwhichcellsfirstquicklystretch,butthengraduallyslidebyoneanother).Todistinguishbetweenthesealternatives,wehaveexaminedcellshapesincentrifugallydeformedchickliveraggregates.Lightandelectronmicrographsshowthatinitiallystretchedcellswithinflatteningaggregatesgraduallyreassumetheiroriginalundisturbedshapesduringprolongedcentrifugation.Therefore,althoughthecellsthemselvesmayreacttostretchingforceslikeelasticsolids,theyslowlyslippastoneanotherwithinaggregatestorelaxstretchingforces,endowingtheaggregateswithliquidproperties.Slowcellslippagecanaccountnotonlyfortemporaryelasticsolidityandviscousliquidflowincellaggregates,butalsoforstress-freechangesinthepositionsandconfigurationsoftissuesmigratingwithindevelopingembryos.Regulationofcellslippagepropertiesmayshiftthemorphogeneticdependenceofembryonictissuesinsuccessivedevelopmentalstagesfromintracellulartointercellularforce-generatingmechanisms,orviceversa.
