Stored and polysomal ribosomes of mouse ova☆
[摘要] RNPparticlesofovulatedmouseova,labeledbyexposureofgrowingoocytesto[3H]uridine,weredisplayedonsucrosegradients.Understandardsaltconditions,radioactivitywasobservedcoincidingwithliverribosomalsubunits,monomers,andpolysomes.TheRNAfromeachregionofthegradientwasisolatedandwasfoundtocontaintheexpectedspeciesoflabeled18Sand/or28SribosomalRNA.HeterogeneousRNPparticleswerewidelydistributedinthegradient.FromdataonRNasesensitivityandresistancetodissociationinhighsalt,itwasestimatedthat20–25%ofthetotalribosomeswereinpolysomes.Nodifferenceinthedistributionwasobservedwhenribosomeswerelabeledintheearlyorlategrowthphaseoftheoocyte.Theevidencesuggestedthatthenonpolysomalsubunitsandmonomerswereunabletoformahighsalt-stablecomplexinthepresenceofpoly(U)andfactorsforproteinsynthesis.Thus,thebulkoftheribosomesareinactiveinproteinsynthesisinovulatedovaandareapparentlystoredforuseinembryonicdevelopment.
