Mitochondrial development during myogenesis☆
[摘要] Theenzymaticandultrastructuralpatternofmitochondrialdifferentiationwasinvestigatedduringmyogenesis.SuccinatecytochromeCreductase(SCR),amitochondrialenzymecomplex,increasedinactivityindevelopingchickthighmuscleinvivoandinvitro.SCRincreaseinvitrooccurredsubsequenttomyoblastfusionandcorrelatedwiththeperiodofincreasingcreatinephosphokinase(CPK)activity.Fusion-arrestandCa2+-reversalexperimentsindicatedanapparentcoordinationbetweenCPKandSCRenzymaticincreasesandfusion.AnalysisofSCRactivityinfibroblastculturessuggestedthattheenzymaticincreasesobservedindifferentiatingmuscleculturesreflectedmyocytedifferentiation,ratherthanfibroblastcontaminationorauniquepropertyofthetissuecultureenvironment.MorphologicaltransitionsinthemyogenicmitochondriaweretemporallycorrelatedwithincreasedSCRactivity.Duringmyogenesis,themitochondriaenlargedinlengthandvolume,exhibitedanincreaseinmatrixdensity,orientedinparalleltothelongaxisofthemyofibrils,andcontainedincreasednumbersofparallelcristae.Manymitochondriainfusion-inhibitedmuscleculturesresembledthosefoundinprefusionmyoblasts,althoughmaturemitochondriawereobservedinsomefusion-blockedcells.Quantitativestereologicalanalysesofthesemitochondrialchangesparallelthebiochemicaldataandsuggestthatultrastructuralandenzymaticchangesinthemitochondriaareanintegralpartofmyodifferentiation.
