Hormonal regulation of epidermis-specific protein and messenger RNA synthesis in amphibian metamorphosis
[摘要] Experimentsusingamonospecificantibodydirectedagainstonetypeofepidermis-specifickeratinfromadultskinoftheamphibianXenopuslaevishavedemonstratedthatpolysomessynthesizingthisproteinfirstappearwithinlarvalskinduringnaturalmetamorphosis.Furtherexperimentsdemonstratedthatthesynthesisofkeratinwithinlarvalskincouldbeinducedprecociouslybythethyroidhormone,3,3′,5-triiodo-l-thyronine,bothinvivoandwhentheisolatedlarvalskinisculturedinvitro.TheearliestdevelopmentalageresponsivetosuchhormoneinductionappearedtobeStage5052oflarvaldevelopment.Thisisabout20–24daysbeforekeratinwouldnormallymakeitsappearancewithintheskinduringnaturalmetamorphosis.HormonetreatmentoftadpolesatthisagewillalsocauseaprecociousincreaseintheamountofkeratinmessengerRNApresentwithinlarvalskin.Thishasbeendemonstrateddirectlybytheisolationofpoly(A)-containingmessengerRNAfromhormone-treatedlarvaeanditstranslationinawheatgermcell-freesystemtogiveimmunoprecipitablekeratin.Peptideanalysisoftheinvitrotranslationproductindicatesthatthehormone-inducedmRNAprobablycodesforaninitialproteinproductthatisslightlylargerthankeratinitself.
