Interaction between glutathione and the endoplasmic reticulum in cyclic protein synthesis in sea urchin embryos☆
[摘要] Interactionbetweenanoxidoreductionsystemandcyclicproteinsynthesiswasstudiedinseaurchinembryos.Whenassayedenzymatically,inbothinvivoandinvitrosystems,thecontentsofGSHandGSSGvariedinverselyinacyclicfashion.Diamideat0.5mMinhibitedaminoacidincorporationinnotonlythecyclicphasebutalsothebasalphase,but4-nitroquinoline-N-oxideat1μMinhibitedonlythecyclicphase.Seaurchinembryoscontainedmembrane-boundribosomes,andpulse-labelingwithaminoacidssuggestedthatfreeribosomeswereresponsibleforthebasalphaseandmembrane-boundribosomeswereresponsibleforthecyclicphaseofaminoacidincorporation.Thiol-disulfideinterchangingenzymewasfoundintheendoplasmicreticulumfraction.Anextractoftheendoplasmicreticulmcausedstimulationofbindingofacetylphenylalanyl-tRNAto40SribosomesandpolyphenylalaninesynthesisinthepresenceoflowGSHconcentrations.AnextractoftheendoplasmicreticulumalsocatalyzedoxidoreductionfromGSHtotheKCl-solubleprotein.Thus,theperiodicstimulationofproteinsynthesisisinterpretedtobetheresultoftheperiodicactivationofmembrane-boundribosomesbythethiol-disulfideinterchangingenzymewhichacceptsselectivelythesignalfromtheGSHcycle.
