Changes in rate of RNA synthesis and ribosomal gene number during oogenesis of Drosophila melanogaster☆
[摘要] AnewmethodforseparatingDrosophilaeggchambersintodifferentdevelopmentalclasses(Jacobs-LorenaandCrippa,1977)madeitpossibletostudychangesintherateofribosomalRNA(rRNA),5SRNA,andtRNAsynthesisandthechangesinribosomalgenenumberduringoogenesis.SynthesisofRNAwasmeasuredby[3H]uridineincorporationinvivoandsubsequentanalysisonsucrosegradientsorgelelectrophoresis.Specificradioactivityofnucleotidepoolshasalsobeendetermined.RibosomalgenenumberhasbeenmeasuredbyhybridizationofeggchamberDNAtorRNAofhighspecificradioactivity.OurfindingsledustoconcludethatinDrosophilamelanogaster:(i)rRNA,5SRNA,andtRNAaresynthesizedinallstagesofoogenesis.(ii)Ineverystage,rRNAisthemainRNAspeciessynthesized.(iii)TherateofrRNA,5SRNA,andtRNAsynthesisincreasesgreatlyduringoogenesisandisparalleledbyasimilarincreaseinribosomalgenenumberresultingfromthepolyploidizationofthenursecellnuclei.
