Starfish oocyte maturation and fertilization: Intracellular pH is not involved in activation☆
[摘要] IntracellularpH(pHi)wasassayedduringthehormonallyinducedmaturationofoocytesofthestarfishPisasterochraceus.CytoplasmicpHwasmeasuredbytheDMOmethod,andconcurrently,theinitiationofmaturationwasdeterminedbygerminalvesiclebreakdown(GVBD)andtheincreaseinproteinsynthesis(percentageincorporationofaminoacids).Ourresultsindicatethat(1)oocytepHirisesslightlyafterinitiationofmaturation;(2)GVBDisnotinhibitedbyacidifyingpHi;and(3)aminoacidincorporationcanbeaffectedbylargechangesinpHi,butnotbythesmallpHichangepromotedbymaturation.Therefore,activationofGVBDandaminoacidincorporationmustproceedbymechanismswhichdonotincludechangingpHi.Theseconclusionsappeartobetrueofoocyteactivationbyfertilizationaswell,forthepHichangefollowinginseminationisevensmallerthanduringmaturation.Theseresultsarediscussedintermsofmechanismsbywhichdormancyiscontrolled.
