[摘要] Thedistributionofpeanutagglutinin(PNA)bindingsitesinimaginaldiscsisdescribedusingfluorescenceandelectronmicroscopy.PNAbindspreferentiallytothephotoreceptorcellprecursorsineyediscsresultinginarectilineararrayoffluorescentspotsthatreflectsthatlattice-likearrangementofthepresumptiveommatidia.Thelectinbindstotheapicalsurfaceoffixeddisccellsandistakenupinpresumedendocytoticvesiclesinlivingdiscs.Photoreceptorprecursorscanbevisualizedwithfluoresceinisothyocyanate-PNAfromthetimetheyfirstformpreclustersinthemorphogeneticfurrowandthistechniqueisusedtodemonstrateatemperature-sensitivedefectinpreclusterformationinthemutantshibire.PNAislocalizedalongthesidesofmicrovilliofdisccells,ingeneral.ThepreferentialbindingofPNAtophotoreceptorprecursorsisrelatedinparttothehighdensityofapicalmicrovillionthesecells.
