The formation of the pupal cuticle by Drosophila imaginal discs in vitro☆
[摘要] Mass-isolatedimaginaldiscsofDrosophilamelanogasterformachitin-containingpupalprocuticleInvitro.Optimalprocuticledepositionoccurswhenthediscsareincubatedfor4–6hrwith0.5–1.0μg/mlof20-hydroxyecdysoneandthenwithlessthan0.05μg/mlof20-hydroxyecdysone.Theformationofthechitin-containingprocuticleisdemonstratedusingthreeindependentassays:withfluorescene-conjugatedcuticleproteinsthatbindtochitin;byelectronmicroscopy;byincorporationof[3H]glucosamineintoachitinfraction.Synthesisanddepositionofpupalcuticleproteinsarealsodemonstrated.Incorporationof[3H]glucosamineintochitinissensitivetoinhibitorsofprotein,RNAandchitinsynthesis,buthaslittlesensitivitytoinhibitorsofDNAsynthesis,anddolichol-dependentglycosylation.
