Ontogeny of hematopoietic cells in Rana pipiens: Precursor cell migration during embryogenesis☆
[摘要] TheinitialmigratorypatternsofhematopoieticprecursorcellsduringearlydevelopmentinRanapipienswereinvestigatedbytracingthedistributionofdiploidand/ortriploidcellstransplantedtocytogeneticallydistinctembryos.Oneexperimentalseriesinvolvedtheproductionofanterior/posteriorchimerasatthelevelofthepronephrosduringembryonic(Shumway)stages14(62hr),15(67hr),16(72hr),and17(84hr).MicrodensitometricFeulgen-DNAmeasurementsoflarvalhematopoieticorgansharvestedbetween25and30dayspostfertilizationindicatedthatthemajorityofprecursorcellsarelocatedposteriortothepronephrosatstage14.Thesedataalsosuggestthatbetweenstages15and17thereisaposteriortoanteriormigration.Reciprocaltransplantationofcytogeneticallylabeleddorsallateralplatemesodermatstages14and15,followedbyF-DNAanalysisofthedorsalaortaeandpronephroses-posteriorcardinalveinsatembryonicstage20,demonstratedthatprecursorcellsderivedfromthetransplantedregioninvadethedevelopingcirculatorynetworkduringthistimeperiod.Observationofcellsfromstage20pronephrosesandaortaestainedwithWright-Giemsasuggeststhattheseprecursorcellsaremorphologicallyprimitiveatthetimeofinitialcolonization.Therefore,weproposethathematopoieticprecursorcells,possiblyderivedfromlateralplatemesoderm,migrateinterstitiallyfromtheposterioroftheembryoduringearlydevelopmentandinvadetheanteriorlylocatedpronephrosesanddorsalaortaeduringstage20.
