A gene product of the mouse t complex with chemical properties of a cell surface-associated component of the extracellular matrix☆

[摘要] p63/6.9isamajortesticularcellproteincodedforbyagene,calledTcp-1,withinthemousetcomplex.Allwild-typechromosomescarrytheTcp-1ballelewhichcodesforabasicformofthisprotein,whileallcompletemutantthaplotypescarrytheTcp-1aallelewhichcodesforanacidicformofthisprotein.GeneticstudieshavedemonstratedacorrelationbetweentheTcp-1geneandcertainthaplotypeeffectsonspermdifferentiationandmaturation.Inthisreport,aninitialbiochemicalanalysisofthep63/6.9proteinispresented.Thedataprovideevidencethatp63/6.9iscloselyassociatedwiththeexternalsurfaceoftesticularcellsbutnotasanintegralmembranecomponent.Somepropertiesofthetesticularformofthistcomplexgeneproductaresimilartothosereportedforthematrixproteinsfibronectinandlaminin.Thepossibilityissuggestedthatprimaryeffectsofthaplotypesonspermdifferentiationcouldbeexertedthroughtheextracellularmatrix.p63/6.9isalsopresentatalowerlevelwithinthecytoplasmandmembranesofF9teratocarcinomacells.Itappearsthatthelevelofp63/6.9synthesisandtheexactnatureofp63/6.9intra-and/orintermolecularinteractionsareundertissue-specificcontrol.
