Evidence for the guidance of pronephric duct migration by a craniocaudally traveling adhesion gradient☆☆☆
[摘要] TheseexperimentswereundertakentoidentifythegeneralnatureofthemechanismthatguidesthemigrationoftheAmbystomapronephricductalongtheventraledgeofthesomitefilefromitsanteriororigintothecloaca.Usingscanningelectronmicroscopyinconjunctionwithmicrosurgery,wehavesoughttodistinguishamongsuchpossibilitiesaschemotaxis,contactguidance,andgradientsofadhesivenesstothesubstratum.Apronephricductprimordiumtransplantedtotheflankofahostventraltotheprimaryductmigratesdorsocaudallyacrosstheflanktofusewiththeprimaryduct.Removalofpotentialsourcesofdistantattractiondoesnotalterthisbehavior,nordomigratingsecondaryductsfollowanyvisiblestructures.Avarietyoftransplantationexperimentsrevealthattheguidanceinformationisnotonlyorientedbutalsodirectionallypolarizedandtravelscaudadasawave.Theseresultsmilitateagainstchemotaxisandcontactguidanceasguidinginfluencesandindicatethatthecellsofthepronephricducttiparedirectedintheirmigrationbylocalinformationwhichpassescaudadovertheduct'smesodermalsubstratumasawaveinregisterwiththeadvancingwaveofsomitesegmentation.Weproposethatthisduct-guidinginformationmaybeatravelinggradientofflankmesodermcelladhesiveness.
