Developmental lesions in the Agametic mutant of Drosophila melanogaster
[摘要] Agametic,amaternal-effectmutation,causestheabsenceofgermcellsinapproximately40%ofthegonadsoffliesderivedfromhomozygousfemales.Thenatureofthedeficiencyintheeggsproducedbytheseflieswasexamined.Ultrastructuralabnormalitieswereseeninthepolargranulesofsomeeggsshortlyafterfertilization.Althoughanormalnumberofpolecellsform,someareabnormalwithdegeneratingpolargranulesandnuclearbodiesandtheycontainmyeloidbodies.Thepolecellsreachthegonadsandat14hrofdevelopmentallthegonadscontaingermcells.However,in40%ofthegonadsthegermcellsbecomenecroticanddisappear.Thus,thesourceofagameticgonadsintheadultisembryonicdeathofpolecellsinsomegonads.Totestwhetherthisgonadaldeathisanautonomousdeficiencyofthemutantpolecells,mosaicpolecellpopulationswereproducedbyreciprocalpolecelltransplantation.Inbothtypesoftransplants,themutantpolecellsdiedautonomously.Ineightinstancesgonadscontainingonlydonorpolecellswereobtained.Sincemutantpolecellsdiewhenwild-typepolecellsnormallybegindividing,wesuggestthatthelesionaffectstheabilityofthesemutantpolecellstoreenterthecellcycle.
