Spontaneous release of nucleosome cores from embryoblast chromatin☆
[摘要] Duringinvitroincubationoftherabbitblastocyst,chromatinfragmentsofthenucleosomeseriesarereleasedspontaneouslybycellsofthepluripotentembryoblastbutnotbythedevelopmentallyrestrictedtrophoblast.TheDNAofthesefragmentsisdoublestrandedandlinear,andrepresentstotalgenomicDNA.Thefragmentsarestableforatleast24hrandareinitiallyfoundinthecytoplasmofintactcells.ThekineticsofradiolabelingoftheDNAcomponentofthesefragmentssuggestthattheyoriginatefromactivelyreplicatingcells,possiblyonlyatthetimeofmitosis.DNaseII-likeactivity,oneoftheendonucleasesknownpreferentiallytoattackspacerDNAbetweennucleosomecores,isreadilydetectableintheblastocyst,althoughtheevidenceforitsinvolvementintheobservedchromatinfragmentationremainscircumstantial.Thesenonrandomchromatinbreaks,presumablyrepresentinganearlyeventincelldeath,occurmorefrequentlyundersuboptimalconditionsofincubationbutarestilldetectableinembryosdevelopingnormally.Thereisnoevidenceforlysosomalactivationinthesedyingcells.Thesefindings,togetherwithpreviousreportsofidenticalchromatinfragmentationinothercellsystems,suggestthatthisformofcelldeathintheembryoblastmaybeonlyonepossibleresultofanactiveprocess,andthatthemechanismsresponsibleforgeneratingdouble-strandbreaksininternucleosomalDNAmaybeofmorebiologicalsignificancethanDNAdegradationalone.
