Postnatal maturation of nerve-muscle junctions in hindlimb muscles of the mouse
[摘要] Themorphologicalmaturationofnerve-musclejunctionsinhindlimbmusclesofthemousewasstudiedduringthefirstmonthafterbirth.Atbirththeadultpopulationofmusclefibersandmotoraxonsisnearlycompleteandthelatterareabouttobecomemyelinatedintheirextramuscularregion.Eachneonatalmusclefiberisinnervatedwithinawell-definedjunctionalzonebynumerousfineprocessesofseveralmotoraxons.Thiszoneisoccupiedroughlyuniformlybyahighdensityofacetylcholinereceptors(AChR,labeledwithafluorescentconjugateofα-bungarotoxin).Polyneuronalinnervationofindividualmusclefibersislostduringthesecondweekafterbirth.Followingtheeliminationofredundantsynapticcontactsthesurvivingpreterminalaxonsgrowandbecomemyelinatedandtheirterminalsexpandwithinthejunctionalzonewhichbeginstogrowmorerapidly.ThedistributionofAChRchangesmarkedlyduringthethirdweekafterbirth.SpotsoflowAChRdensityappearwhichenlargetoformthenonsynapticregionsofthematurejunctionalzoneasthereceptorsbecomecloselyassociatedwiththematuringbranchesofthemotoraxonterminals.Thesubsequentincreaseinareaofthejunctionalzonecloselyparallelsthatofthemusclefiberasawhole.
