Ecdysteroid titers during pupal and adult development in Drosophila melanogaster☆
[摘要] Ecdysteroidtitersweredeterminedbyradioimmunoassayduringprepupal,pupal,andadultdevelopment,andduringadulthoodinDrosophilamelanogastermalesandfemalessynchronizedatpupariationoradulteclosion.Atseveraltimesindevelopmentecdysteroidswereidentifiedusingchromatographictechniques.Afteranecdysteroidpeakatpupariation,asmallpeakoccurredafter10hrat25°C,withasubsequentrapidandlargeincreaseinactivityat30hr.One,andpossiblytwomorerapidpeaksoccurredduringthemid-pupalperiod,withagradualdeclinetobasallevelsinthepharateadult.Duringthefirstdayofadulthood,theecdysteroidleveldeclinedfurtherandremainedatapproximately10pg/mgfreshweightfor2weeks.Titerswerenotsignificantlydifferentbetweenmalesandfemalesatanyofthestagestested,withthebiologicallyactive20-hydroxyecdysonehormoneaccountingfor70to90%oftheecdysteroidsinbothsexes.Ecdysteroidsinadultfemaleswerelocalizedtothehemolymphandabdominaltissue,butnottotheovaries.Itisconcludedthatdifferencesintheconcentrationof20-hydroxyecdysoneinmalesandfemalesarenotresponsibleforthesexuallydimorphicexpressionoftheyolkproteingenes,andthatasourceotherthantheovariesprovideshormoneinfemalesforthispurpose.
