Mitogenic property of the apical ectodermal ridge☆
[摘要] Whiletheapicalectodermalridge(AER)iswellknownforitsrequiredroleinthedevelopmentofdistalpartsofthelimbandforitsabilitytostimulatelimbduplications,themechanismofitsactionisunknown.InthisstudyweuseaculturesystempreviouslydevelopedbyM.GlobusandS.Vethamany-Globus(1976,Differentiation6,91–96)inwhichanAERgraftedontoahigh-densitycellcultureoflimbmesenchymestimulatestheformationofanoutgrowth.Time-lapsemoviestakenduringtheoutgrowthperioddemonstratednocellularactivitiesotherthancelldivision.BoththemitoticindexandlabelingindexinthemesenchymeweresignificantlyelevatedundertheAERascomparedtothatwithoutAER,indicatingthattheAERprovidesagrowth-promotingstimuluswhichincreasestheproportionofdividingcells.Ontheotherhand,nonridgeectodermhadnodetectableeffectonthemitoticindex.TreatmentofcultureswithcytosinearabinosidebothinhibitedDNAsynthesisandpreventedAER-inducedoutgrowth.TheseresultsdemonstrateamitogeniccapacityofAERtissueandsuggestthatmesenchymaloutgrowthrequiresthisactivity.ThemitogenicpropertyoftheAERisconsideredinrelationtolimboutgrowthinsitu.
