Inhibition of myoblast fusion by lysosomotropic amines☆
[摘要] Fusionofculturedchickembryomyoblastswasinhibitedbytreatmentwithseverallysosomotropicamines.Theconcentrationsrequiredforhalf-maximalinhibitionoffusionwereapproximately2μMforchloroquine,30μMfortributylamine,3.2mMforammoniumchloride,and3.3mMformethylamine.Alltheaminesinhibitedfusionappreciablyatconcentrationslowerthanthosethatreducedcelldensity.Boththerateandextentoffusionwereaffectedbytheamines,whichhadtobepresentforabout20hrbeforetheusualonsetoffusion.Inhibitionoffusionwasreversiblebytransferofinhibitedcellstofreshmedium.Theaminesdidnotcauseaccumulationofanondialyzableinhibitorintheculturemedium.Levelsofcreatinekinaseincreasedbyeight-foldormorebetween18and65hrinculturestreatedwithtributylamineorchloroquine,althoughthisincreasewasnotaspronouncedasincontrolcultures.Theincreasedcreatinekinaseactivityinamine-treatedcultureswasduemainlytotheBBandMBisozymes,withrelativelylittleincreaseintheMMisozyme.
