Integration of sperm and egg plasma membrane components at fertilization☆
[摘要] Studiesexaminingtheintegrationofthespermandeggplasmamembranes,subsequenttogametefusioninthesurfclam,Spisulasolidissima,werecarriedoutemployingtheconcanavalinA-horseradishperoxidase-diaminobenzidineprocedure(ConA-HRP-DAB).WhenunfertilizedSpisulaeggswereincubatedinConA,eitherpriortoorafteraldehydefixationandreactedwithHRP-DAB,enzymaticprecipitatewasfoundassociatedwiththevitellinelayerandplasmalemma.Theplasmamembranesofspermtreatedinasimilarmannerfailedtostain.TheplasmamembranesoffertilizedeggsreactedwithConA-HRP-DABandexaminedby1minpostinseminationwereassociateduniformlywithenzymaticprecipitateexceptatsitesofspermincorporation.Theseportionsofunstainedplasmamembranewerederivedfromthespermatozoonanddelimitedthecontentsofthefertilizationcone.From2to4minpostinsemination,HRP-DABreactionproductbecameassociatedwiththeplasmamembranedelimitingthefertilizationcone.By4minpostinseminationnodifferenceinstainingoftheplasmamembranesderivedfromtheeggorthesperm(plasmalemmadelimitingthefertilizationcone)wasdetected.EvidenceispresentedsuggestingthattheacquisitionofHRP-DABreactionproductbytheformerspermplasmalemmaisduetothemovementofConAbindingsitesfromtheeggplasmamembrane.
