Cell-specific regulation of protein synthesis in the sea urchin gastrula: A two-dimensional electrophoretic study☆
[摘要] Proteinsynthesisinthetwocelltypesofechinoidearlygastrulaewasanalyzedbytwo-dimensionalpolyacrylamidegelelectrophoresisandfluorography.EpithelialcellsandprimarymesenchymecellswereisolatedfromearlygastrulaeasdescribedbyM.A.HarkeyandA.H.Whiteley,1980(Wilhelm.Roux's.Arch.189,111–112).Newlysynthesizedproteinswerelabeledwith[3H]valine,extractedinSDSbuffer,andanalyzedelectrophoretically.Ofthe454labeledproteinsanalyzed,58incorporated[3H]valineatmarkedlydifferentrelativeratesinthetwocelltypes,and69werelabeledexclusivelyinoneortheothercelltype.Themostrapidlysynthesizedproteinsingastrulacellsconstitutedaclasswhichexhibitedamuchhigherdegreeofcellspecificitythanthetotalproteinpopulation.Severaloftheserapidlysynthesizedproteinswereanalyzedindividually.Amongthosethatweresynthesizedpreferentiallyinprimarymesenchymecells,twolow-molecular-weight,acidicproteins,designatedPM28andPM32,accountedfor9–14%ofthetotalproteinsynthesisinprimarymesenchymecellsbutwerebarelydetectableinepithelialcells.Thoseproteinsthatweresynthesizedpreferentiallyintheepithelialcellsincludedseverallow-molecular-weightspecies,probablyhistones,andthecytoskeletalproteins,actinandtubulin.Thesedataindicatethattheprimarymesenchymeandepitheliumoftheearlygastruladifferprofoundlywithrespecttothesynthesisofspecificproteins.
